I have PosterShop Pro 3.5 Version. How can I use My Mimaki CJ121 Plotter with this version?

I think you mean the Mimaki CG121 not the CJ121. This model is a cutting plotter not a printing plotter. I think Onyx didn't release a Vinyl cutting software in order to be used with a equipment as the Mimaki CG-121

You can use software as CASmate or Inspire from Scanvec-Amiable, or Flexisign of the same house, etc.
